The Cancer Coessentiality Network!
Genome-scale knockdown (shRNA) and knockout (CRISPR) screens in human cell lines tell us which genes cause fitness defects when they are disrupted. Systematic knockout screens in yeast have shown us that genes with correlated fitness profiles across a panel of diverse genetic backgrounds are often involved in the same biological processes. We show that the same is true in human cells: genes with correlated fitness profiles across 112 shRNA screens in cancer cell lines are more than 30-fold enriched for cofunctionality. Moreover, clusters of correlated genes can indicate the specific genetic backgrounds in which they are essential, highlighting both tumor-specific genetic vulnerabilities and their cognate biomarkers.
